Method

A narrative review of the current literature about Histamine and migraine was conducted.

Result

Histamine presents a significant role in migraine pathogenesis: supporting the neurogenic inflammation pathway. The interaction between calcitonin-gene related protein (CGRP) AND mast cells (MC) appears in trigeminal ganglia (TG) and trigeminal afferents sensitisation. Histamine attaches with differing affinities to four distinct histaminergic G-protein coupled receptors, stimulating protein kinases, or triggering calcium release with the subsequent mode of actions. Allergy and gastric acid secretion frequently managed with the Histamine one receptor (H1R) and Histamine two receptor (H2R) antagonists, respectively but their antagonism is seemingly futile for migraine. H3R serves as a heteroreceptor or as an autoreceptor, decreasing the liberation of Histamine and other neurotransmitters. This is a potential spot for anti-nociception and anti-neurogenic inflammation. To date, numerous small clinical trials employing low dose Histamine or Nα-methylhistamine have expressed prophylactic migraine efficacy, presumably via H3R or other undetermined pathways.

Conclusion

The Histamine system communicates with various regions in the CNS and may hypothetically change the response of migraine. Low dose Histamine may be an assuring choice for migraine treatment.
